17-P019 A Pax3/7:Foxc2 negative feedback loop in the somite modulates multipotent stem cell fates

چکیده
mordial germ cells (PGCs) in mice and axolotls, we propose that the deletion of Nanog in species often employed as experimental models, such as frogs and teleosts (Zebrafish; Danio rerio), has been compensated by the evolution of predetermined germ cells in these organisms. Our data highlights Nanog as a major component of the gene regulatory network that governs early development in the major trunk of chordate evolution.

ical role in regulating their entry into the myogenic programme. We had shown that in the Pax3/7 double mutant the myogenic determination genes, Myf5 and MyoD are not activated, leading to a major deficit in skeletal muscle. Pax3 directly activates Myf5, thus promoting a myogenic cell fate. However, it is essential to maintain a balance between differentiation and renewal of the progenitor cell...
متن کاملNotch regulation of myogenic versus endothelial fates of cells that migrate from the somite to the limb.
Multipotent Pax3-positive (Pax3(+)) cells in the somites give rise to skeletal muscle and to cells of the vasculature. We had previously proposed that this cell-fate choice depends on the equilibrium between Pax3 and Foxc2 expression. In this study, we report that the Notch pathway promotes vascular versus skeletal muscle cell fates. Overactivating the Notch pathway specifically in Pax3(+) prog...
متن کاملA Pax3/Dmrt2/Myf5 Regulatory Cascade Functions at the Onset of Myogenesis
All skeletal muscle progenitor cells in the body derive from the dermomyotome, the dorsal epithelial domain of developing somites. These multipotent stem cells express Pax3, and this expression is maintained in the myogenic lineage where Pax3 plays an important role. Identification of Pax3 targets is therefore important for understanding the mechanisms that underlie the onset of myogenesis. In ...
متن کاملRegulation of dorsal somitic cell fates: BMPs and Noggin control the timing and pattern of myogenic regulator expression.
Previous work has indicated that signals from the neural tube, notochord, and surface ectoderm promote somitic myogenesis. Here, we show that somitic myogenesis is under negative regulation as well; BMP signaling serves to inhibit the activation of MyoD and Myf5 in Pax3-expressing cells. Furthermore, we show that the BMP antagonist Noggin is expressed within the dorsomedial lip of the dermomyot...
متن کاملMicroRNA regulation of the paired-box transcription factor Pax3 confers robustness to developmental timing of myogenesis.
Commitment of progenitors in the dermomyotome to myoblast fate is the first step in establishing the body musculature. Pax3 is a crucial transcription factor, important for skeletal muscle development and expressed in myogenic progenitors in the dermomyotome of developing somites and in migratory muscle progenitors that populate the limb buds. Down-regulation of Pax3 is essential to ignite the ...
